The collision occurred on Delta Way at the junction with Hurricane Road. Gloucestershire Police said the road is closed while forensic teams gather evidence.
The injured man has been taken to Southmead hospital in Bristol.
In a statement on Twitter, Gloucestershire Police Specialist Ops said: “One man has died & another is in a critical condition at Southmead hospital in #Bristol following a road traffic collision in #Brockworth on Delta Way at the jtn with Hurricane Road.
“The road remains CLOSED whilst forensic collision investigation work takes place.”
